Having never made an all maple neck before, does one want to seal the holes for position markers with vinyl sealer or similar prior to installation with superglue?

No need to. Just a tiny dot of glue in the bottom of the hole, press in the markers, let dry and sand flush.

Superglue doesn't seem to discolor maple the way it does spruce. But, like he said, you might want to test your CA on a scrap of the specific maple.

But... maybe add the glue after the dot is in. I used to put the glue in first, until I pressed on a dot one day only to have some CA squeeze out land in my eye. I quickly amended my procedure after that.
